The weekly chart ranks the most popular singles based on U.S. streaming record, sales data and radio airplay.
Earlier, rapper PSY's "Gangnam Style" reached No. 2, where it stayed for seven weeks, but never got to No. 1.
BTS have topped the Billboard 200 albums chart four times, but this is their first No. 1 on the singles chart. That means they have broken their own record set by "On", the lead track from the album "Map of the Soul: 7" released in February, which reached No. 4.
"Boy with Luv" and "Fake Love" previously ranked 8th and 10th respectively.
"Dynamite", the band's first song in English, was released on Aug. 21. The band earlier said that they decided to sing the song in English to reach more global fans and give them strength amid the coronavirus pandemic.
Billboard described the song as "made of positive vibes, energy, hope, love, the purity, everything".
